چکیده

The effect of 4-nitroquinoline 1-oxide on the cell-mediated immune response was tested in C57BL/6 female mice. The carcinogen was administered s.c. either 30 days before or just prior to grafting the female recipients with C57BL/6 male skin. Skin graft survival times were noticeably prolonged in mice that received the carcinogen 30 days before grafting. Administration of 4-nitroquinoline 1-oxide just prior to skin grafting, however, yielded an unexpected result in that this group of mice rejected their grafts much sooner than their controls. The cause of this accelerated rejection is not presently known. The tumors that arose following s.c. implantation of 4-nitroquinoline 1-oxide pellets were moderately to highly immunogeneic and had a short latent period. In this respect they are comparable to tumors induced with the polycyclic hydrocarbon carcinogens.

Protective Immunity in Mice Following Immunization with the Cochleate-Based Subunit Influenza Vaccines

High morbidity and mortality of influenza virus infection makes it an important disease world-wide. Mouse is a very well-studied animal model for this disease with similar manifestation to human disease. It would be desirable to induce mucosal as well as circulating immune responses to obtain protection from infection and to decrease the spread of the virus. Cell mediated immunity (proliferativ...

Detection of in vivo DNA repair synthesis in mouse liver and lung induced by treatment with benzo(a)pyrene or 4-nitroquinoline 1-oxide.

We examined in vivo DNA repair synthesis in liver and lung of A/HeJ mice treated with benzo(a)pyrene (BP) or 4-nitroquinoline 1-oxide. To differentiate between the removal of carcinogen metabolite:DNA adducts due to cell turnover and DNA repair, we measured unscheduled DNA synthesis (UDS) in the nonreplicating DNA fraction. Mice were exposed to bromodeoxyuridine pellets 1 hr prior to carcinogen...

The Effect of Nitric Oxide on Mouse Pre-Implantation Embryo Development and Apoptosis

Clinical studies have shown that in pathological conditions such as endometriosis and reproductive tract infection (male and female) there is an activation status of macrophages that produce large quantities of nitric oxide (NO) in addition to other effector molecules. Large amounts of NO may have embryotoxic roles and produce infertility. This study was designed to evaluate the effect of diffe...

Effects of glycerol on 4-nitroquinoline 1-oxide induced pulmonary tumorigenesis in ddY mice.

The effects of glycerol administration on pulmonary tumorigenesis induced by 4-nitroquinoline 1-oxide (4NQO) were examined in male ddY mice, that were given 5% glycerol solution instead of drinking water after a subcutaneous injection of 4NQO. The incidence of pulmonary tumor-bearing mice and the mean number of induced tumors per mouse were significantly enhanced in mice given glycerol after 4N...
